C1498 cells (1 × 105) were incubated overnight in the presence or absence of PS-341 and TRAIL (1 μg/mL). Cells were then washed thoroughly and resuspended in 1 mL fresh media. Various dilutions of cells were transferred to microtiter plates.

Viable cell numbers were estimated at various time points using an MTS assay based on a standard curve with fixed numbers of C1498. The number in parentheses indicates a theoretical number assuming a 4-fold increase in cell number per day.
