Effect of orthovanadate on human erythroid progenitors

Purified human erythroid progenitors (90% pure as determined by flow cytometry for the markers glycophorin A and CD71) were removed from growth factors (EPO, SCF, and insulin-like growth factor 1) on day 6. The cells were then cultured either with growth factors, or without growth factors but with the selected concentrations of orthovanadate, for 20 hours. The cells were analyzed for evidence of apoptosis with Annexin V–FITC and propidium iodide (PI).
