Lethally irradiated Ly-5.1 mice were transplanted with 103 Sca-1+c-kit+LinBM cells (Ly-5.2+) or their entire expanded progeny (approximately 2 × 106 cells) generated after 1 week of serum-free culture in IL-6, IL-11, G-CSF, SCF, FLK, and TPO. Some recipients of expanded cells were simultaneously injected with 10 or 100 μg AcSDKP. Primary mice were analyzed after 4 months. Secondary recipients, receiving a transplant of primary BM cells (0.5 femur/mouse), were analyzed 10 weeks later. Data shown are the mean ± SEM percentage of Ly-5.2+ cells detected in each blood cell compartment and the percentage of normal femoral cellularity and CFC content compared to 4 age-matched B6.SJL mice.
