The 1 × 104 10-week cultured mast cells were incubated in culture wells containing SCF, retinoids, IL-6, or IL-4, alone or in combination. After 2 weeks, the cells were processed for immunocytochemical staining with an antitryptase or antichymase MoAb. The diameter of tryptase+ cells was measured, as described in “Materials and methods.” At the same time, intracellular histamine concentrations were determined by the radioimmunoassay.
