Colony-forming cells were assayed in MethoCult M3230 medium supplemented with the standard growth factor cocktail in the presence or absence of 106 lymph node cells from C57BL/6-lpr/lpr mice (50% DN T cells) or their 5-fold diluted supernatant. After magnetic depletion of B220+CD4+CD8+ cells, TCRα/β−IgM− splenocytes were electronically sorted as described in Materials and Methods. Data are means ± SEM from triplicate dishes. The experiment was performed with 3-month-old mice.
