Thy/Liv grafts were infected with NL4-3 and viable thymocytes were surface stained for CD3, CD4, and CD8. An aliquot of total cells was depleted of CD3+ and CD8+ cells with magnetic beads and then stained for CD34. From these preparations, 2,000 cells of each of the following subpopulations were sorted: immature intrathymic T-progenitor cells (CD3−CD4+CD8−), mature CD4+(CD3+CD4+CD8−), CD34+ progenitors, or total thymocytes. Lysates were serially diluted and amplified for HIV-1 gag. Shown are copy numbers per 1,000 sorted cells as estimated by endpoint dilution of samples from N mice.
