Groups of mice were vaccinated once with 10¹¹ vp of AdC68gag37 vector and boosted 2 months later with 10⁶ pfu of a vaccinia virus vector expressing Gag. Lymphocytes from lymph nodes, peritoneal lavage, and spleens were tested 12 months after the boost for Gag-specific CD8⁺ cells on stimulation with the gag peptide by intracellular cytokine staining for IFN-γ and TNF-α. Percent frequency of CD8⁺ cells positive for either cytokine over all CD8⁺ cells (rows 1 and 2) and the percent of CD8⁺ cells positive for both cytokines (row 3) are shown.
