Cells were treated with the indicated concentrations of TPO for 15 minutes and then PKC activity was measured in the cytosolic and membrane fractions as described in Materials and Methods. PKC activity is expressed relative to the level present in unstimulated cells (control). The data represent the mean of two determinations from one representative experiment.

Relative levels of c-Mpl expression were determined by flow cytometry as described in Materials and Methods.

For unstimulated cells, the levels of PKC in the membrane and cytosolic fractions of the different UT-7 cell clones ranged from 1.9 to 2.5 U/105 cells and from 14.5 to 22.7 U/105cells, respectively. In CMK cells these levels were 1.7 U/105 cells and 10.1 U/105 cells, respectively.
