Vaccinia truncation analysis (Fig 2) established a limited region between 477-561 where the epitope lies. Computer analysis of the sequence of HCMV pp65 shows 4 relatively high scoring peptides in that region, which generally reflects good binding to the MHC.51 Each peptide was synthesized on the ABI 432 peptide synthesizer (Applied Biosystems), and tested for recognition by 3-3F4 (see Fig 3). Note that peptides at pp65491-500 and pp65495-503 overlap.
