CD34+ MNC (5 × 103) were placed in suspension culture in medium containing serum substitute, or animal serum (12.5% HS + 12.5% BCS), along with the indicated cytokines for 10 days. Cytokine concentrations are as noted in Materials and Methods. After 10 days cells were withdrawn from the cultures and assayed for CFU-Meg. Colony counts shown are mean ± SD of three separate studies, each performed in quadruplicate with cells obtained from a different normal donor. Control cells were plated immediately in (TPO + IL-3) and gave rise to 82 ± 23 colonies when enumerated 10 days after initial seeding.
