CD34+ cells, suspended at 2 × 104 cells/mL cell adhesion medium, were incubated at 4°C for 15 minutes with 5 μg/mL IgG1 or H90 MoAb. Aliquots (100 μL) were seeded in triplicate into HA-coated wells, and, when indicated, GM-CSF, IL-3, or SCF (10 ng/mL) were directly added into the wells. After incubation of 37°C for 15 minutes in contact to immobilized HA, nonadherent cells were removed as described in the Materials and Methods. Adherent cells were seeded in a complete methylcellulose medium supplemented with stimulating factors, as described, and CFU-GM and BFU-E were scored at day 16 (short-term methylcellulose assay). Results were expressed as the percentage of colonies obtained in the input (same cells but without adhesion step), as determined in a parallel experiment. The 100% corresponded to an input of 294 ± 15 CFU, comprising 199 ± 20 BFU-E and 95 ± 10 CFU-GM. All cultures were made in triplicate. Data are the means ± SE of three independent experiments.
