Collagen-coated coverslips were exposed to flowing human blood (shear rate, 65/s) for 5.5 minutes. The blood was supplemented via a mixing device with an anticoagulant (thrombin inhibitor Ro 46-6240) together with P-selectin antibody WAPS12.2 (6 donors) or control antibody pl48 (4 donors) or with buffer (13 donors). The coverslips were washed, fixed, and embedded in Epon resin. Cross sections 1.5 mm from the flow entrance were stained, and thrombus dimensions were measured by computer-assisted morphometry as described in Materials and Methods.
