Cell samples from all three tissues (CoH-1, CoH-2, and CoH-3) were analyzed at each purification step using a semiquantitative HIV-1 Gag DNA PCR per 1 × 104 cells (CoH-3 per 1 × 105 cells). The PCR assay had a sensitivity of detecting one HIV-1 proviral copy in the background of 1 × 104 (or 1 × 105) uninfected cells.
p24 ELISA of apheresis samples (diluted 4-fold in shipping medium) or tissue culture medium obtained after the last transduction. The detection limit was 5 pg/mL. All transduction conditions were tested.
