Memory B cells (CD3CD19+CD20+CD27+CD43), CD3CD19+CD20+CD27+CD43+CD69 B cells, and plasmablasts (CD3CD19+CD20CD43+CD27+) from 3 healthy donors were purified by a combination of magnetic cell isolation and cell sorting (according to the gating strategy shown in Figure 1). The cells were plated at 2.5 × 103 cells per well, incubated at 37°C for 12 h, and analyzed for spontaneous antibody secretion by ELISPOT. ND, not done.
