Memory B cells (CD3⁻CD19⁺CD20⁺CD27⁺CD43⁻), CD3⁻CD19⁺CD20⁺CD27⁺CD43⁺CD69⁻ B cells, and plasmablasts (CD3⁻CD19⁺CD20⁻CD43⁺CD27⁺) from 3 healthy donors were purified by a combination of magnetic cell isolation and cell sorting (according to the gating strategy shown in Figure 1). The cells were plated at 2.5 × 10³ cells per well, incubated at 37°C for 12 h, and analyzed for spontaneous antibody secretion by ELISPOT. ND, not done.
