Cells gated as monocytes were distinguished from lymphocytes by LDS 751 fluorescence, excluding regions of overlap between populations. Substantial overlap between LDS 751 fluorescence of lymphocyte and monocyte populations, as seen in Figure 2, was seen for patients 1 and 2. Recorded cell numbers for patient 5 exclude cells used for mixing with donor cells (Figure 3C-D). Normal monocytes versus monosomy 7 monocytes were classified by gating (supplemental Figure 1). No statistical difference was found between the proportions of monosomy 7 cells identified by conventional cytogenetics versus IC Flow-FISH, as determined by 2-sample χ2 tests for equality of proportions (with continuity corrections) performed in R.
