Noncultured LSK (Ly5.1) cells and Delta1ext-IgG-cultured cells in limiting doses, along with 1 × 105 syngeneic Ly5.1 BM cells, were injected into nonirradiated Rag−/− mice. Successful T-cell reconstitution after HCT was defined as the presence of at least 0.5% donor CD3+ blood T cells at 5 weeks in nonirradiated mice. The data from several limiting dilution experiments were pooled and analyzed by applying Poisson statistics to the single-hit model.
