Analysis of the ability of Lin cells expressing HA to reconstitute hemopoiesis in vivo

Mice were injected with either 5000 to 6000 LinHA+ cells (4 mice) or 500 LinSca+HA+ cells (2 mice) and analyzed 4 to 8 weeks after transplantation. Peripheral blood was collected and the red blood cells lysed using ammonium chloride. The percentage of donor cells was determined using Ly5.1 antibody labeling and was 9% ± 1.7% and 9.4% ± 0.02% following transplantation of 5000 to 6000 LinHA+ cells or 500 LinSca+HA+ cells, respectively. White blood cells were labeled for macrophages and granulocytes (Mac-1/Gr-1), B cells (B220), and T cells (CD4/CD8) and the proportion of donor analyzed using an Ly5.1 antibody.
